Leksum Inks Agreement with Medical University of Vienna for LK-ICU Clinical Trial Preparatory Work

Leksum LLC, an early-stage life sciences company based in Boston, Massachusetts, has signed an agreement with the Department of Anesthesia, Intensive Care Medicine and Pain Medicine at the Medical University of Vienna, with Phoenestra GmbH as a co-signatory, to conduct preparatory work leading up to Phase I/II first-in-human clinical trials for its investigational therapy LK-ICU. The agreement represents a critical step in advancing a potential treatment for ICU-acquired weakness, a condition that can prolong mechanical ventilation and significantly impair patient quality of life.

Preclinical studies of LK-ICU, which consists of extracellular vesicles derived from bone marrow mesenchymal stromal cells, indicate an almost complete preservation of both diaphragm muscle fiber size and function that is normally lost with the use of mechanical ventilation. This finding is particularly important because diaphragm weakness is a major contributor to difficult weaning from ventilators and extended stays in intensive care. Extended weaning from mechanical ventilation, often resulting from ICU-acquired weakness, is estimated to impact the quality-of-life of hundreds of thousands of patients a year in the U.S. and Europe. By preserving diaphragm muscle integrity, LK-ICU could potentially reduce the duration of mechanical ventilation, lower healthcare costs, and improve long-term outcomes for critically ill patients.

The agreement with the Medical University of Vienna, a leading European academic medical center, provides Leksum with access to clinical expertise and infrastructure necessary for planning first-in-human trials. Leksum plans to start efforts under the agreement upon closing its Series A financing round, expects the planning phase to last several months, and intends to use European clinical data to support its U.S. regulatory path. This transatlantic strategy could accelerate the development timeline and facilitate eventual market approval in both regions. For more details, click here.

Leksum is developing advanced biologic therapies to address acquired muscle myopathies. In addition to LK-ICU, the company's pipeline includes skeletal muscle conditions such as sarcopenia, cancer cachexia, and muscle loss due to the use of weight-loss drugs (i.e., GLP-1s). The successful development of LK-ICU could therefore validate a broader platform for treating muscle-wasting conditions, with implications for millions of patients worldwide. As Leksum advances its programs, the medical community will be watching closely to see whether this novel extracellular vesicle approach can translate from promising preclinical results to effective clinical therapies.
